Een voedselcrisis vereist een strakke organisatie en snelle analyse-apparatuuur.
Het Wageningse RIKILT gebruikt UPLC en tweedimensionale GC om snel gevaarlijke stoffen op te sporen. Calamiteitenleider Wim Traag: ‘Eigenlijk moet het werk gisteren af zijn.’
“In tijden van crisis ligt het gewone werk even stil, dan is het alle hens aan dek.” Aan het woord is Wim Traag, leider van het calamiteitenonderzoek bij het RIKILT – Instituut voor Voedselveiligheid in Wageningen. Dit instituut analyseert voedsel dat misschien gevaarlijk is voor de consument en veevoer dat mogelijk besmet is.
“Als er vreemde dingen gebeuren met dieren of mensen, dan komt het RIKILT in actie”, vertelt Traag. Hij zorgt ervoor dat de apparaten, chemicaliën en mensen aanwezig zijn om alle analyses uit te voeren. “We halen collega’s van andere onderdelen van Wageningen UR erbij. Dat zijn getrainde analisten, die onze systemen goed kennen. Het werk gaat veertien uur per dag door, weken lang, soms wel drie maanden.”
Eén telefoontje van het ministerie van Landbouw zet de machinerie in werking. Het RIKILT heeft hiervoor een speciaal nummer dat 24 uur per dag bereikbaar is. Het voert bij de eerste verdachte monsters een total scan uit, met chromatografie onder ultrahoge druk (UPLC) en 2D-GC.
Detectie gaat met TOF-MS. Traag: “We moeten gewoon een monster in het systeem kunnen stoppen en kijken of er giftige stoffen in zitten.” Hiervoor is er een uitgebreide bibliotheek met referentiespectra. Voor de GC bestaat die uit 360 veelvoorkomende bestrijdingsmiddelen. Mocht er geen hit zijn binnen die bibliotheek, dan heeft RIKILT nog een grotere bibliotheek met duizenden pesticiden.
Afbiggen
“We denken dat de total scan een grote vlucht gaat nemen”, vertelt Traag. “De problemen waarmee we te maken krijgen worden namelijk ook steeds breder. Vroeger kreeg je vragen als: ‘Kijk of stof X hierin zit’. Nu is de vraagstelling veel ruimer. Boeren vragen bijvoorbeeld waarom hun zeugen niet kunnen afbiggen of waarom er zoveel dode kraaien op hun weiland liggen.”
Als er een vermoeden is welk soort stof het probleem veroorzaakt, dan wordt er specifieker getest. “Bij dat biggenprobleem duidden de symptomen op het gebruik van hormonen. We hebben toen monsters van de voeding door een bioassay gehaald die hormonen detecteert. Toen die test afging, hebben we het voer uitgebreid gescreend. Het bleek besmet te zijn met medroxyprogesteronacetaat.”
Vaak heeft RIKILT ook informatie van de klant nodig, zoals bij het geval van de dode kraaien. Traag: “Maar de klant gaat niet altijd met de billen bloot, die houdt het soms expres vaag. Dan duurt de analyse echter ook langer.” Komt het RIKILT iets tegen dat niet in de haak is, dan heeft het instituut wel de plicht om dat te melden bij de betreffende overheid.
Nadat de inhoud van de eerste monsters is vastgesteld, neemt het ministerie van Landbouw monsters bij andere bedrijven die mogelijk ook getroffen zijn. Die komen dan met tientallen tot honderden tegelijk binnen. Haast is geboden, want zolang er geen uitslag is, mogen de verdachte boerderijen geen melk of vee afvoeren, met alle economische gevolgen van dien. Traag: “Voor dioxine hebben we vier dagen, voor zware metalen 24 uur. Maar eigenlijk moet het altijd gisteren af zijn.”
Toxines
Het UPLC-systeem dat het RIKILT voor analyses gebruikt, levert in elk geval veel tijdwinst op. “Bij UPLC kun je de flowrate verhogen zonder hierbij resolutie en piekcapaciteit te verliezen, de analysetijd kan soms wel tot een factor tien worden gereduceerd”, vertelt Arjen Gerssen, aio bij het RIKILT. Voor zijn onderzoek naar diarreeveroorzakende toxines in schelpdieren is hij vaak op de UPLC aan het werk. “Verder is ook de injectietechniek verbeterd. De vloeistof gaat na opzuigen in één keer door naar de kolom, waardoor de injectie sneller en beter reproduceerbaar is dan bij oude autosamplers.”
Voor routinematige methoden is UPLC echter niet altijd geschikt. Gerssen: “Een UPLC is veel duurder in aanschaf dan een LC. Je moet dus bedenken of je die tijdwinst echt nodig hebt, of dat je met een LC toe kan. Als je overnacht kunt meten, dan maakt het niet uit of je meting om negen uur ’s avonds klaar is of om zes uur ’s morgens de volgende dag.”
Het RIKILT analyseert schelpdieren op dit moment dan ook met LC-MS/MS op de aanwezigheid van toxines. “De mosselen komen hier binnen op donderdagochtend. We hebben dan tot vrijdagochtend twaalf uur de tijd om te kijken of er toxines inzitten. Is de batch in orde, dan gaan ze de handel in. Deze analyse kan best met LC.”
Ook ziet Gerssen andere nadelen van UPLC. “Doordat de deeltjesgrootte in de kolom kleiner is dan bij conventionele LC is er een groter risico op verstopping. Dat stelt hogere eisen aan de monstervoorbewerking. Als er toch een verstopping optreedt, is de kolom ook vaak meteen kaduuk.”
Het systeem kan alleen met de computer worden bediend. “Als er een hardwarefout is bij de autosampler, dan is het hele systeem meteen onbruikbaar. Omdat het een gesealed systeem is, moet je bij problemen meteen de monteur laten komen. Lekker zelf dingen uit elkaar schroeven is er niet meer bij.”
Calamiteitencoördinator Traag ziet voor de toekomst wel meer potentie voor UPLC: “Over vijf jaar heeft UPLC de klassieke HPLC verdrongen. Natuurlijk is aanschaf duur, maar als je het systeem eenmaal hebt, moet je het zo veel mogelijk gebruiken. Bovendien wordt het systeem steeds goedkoper.”
Buisjes
Voor analyse van pesticiden in groenten, fruit, bloemen en veevoer gebruikt het
RIKILT een 2D-GC systeem, ook met TOF-MS-detectie. Injectie gaat automatisch. “Met 2D-GC zie je alles. We kunnen meer dan 80 procent van alle referentiestoffen volledig scheiden, en dan maken we nog niet eens gebruik van de unieke massa’s. Een gewone GC scheidt slechts 20 procent”, vertelt RIKILT-medewerker Martijn van der Lee, die onderzoek doet naar analysemethoden voor pesticiden en contaminanten. Voordat het monster op de tweede GC-
kolom wordt gebracht, moet het eerst weer worden gecondenseerd. “Dat doen we met vloeibare stikstof, daar gaat per zestig analyses een vat van 120 liter doorheen.”
Omdat er weinig data verloren mogen gaan, is de monstervoorbewerking erg breed. Van der Lee: “Het liefst breng je het monster direct op je GC-kolom, zodat je niets mist. Maar dat is natuurlijk niet mogelijk. Voor plantaardige monsters doen we een korte voorbewerking van een kwartiertje, maar met veevoer ben je wel een uur bezig. Dit soort monsters gaan overnacht. Door onze brede opzet blijft er wel veel vervuiling in het monster achter.”
De glazen linerbuisjes die gebruikt worden bij de injectie zijn het bewijs. In veel buisjes zijn natte stukjes plantenmateriaal te zien. De GC-installatie van het
RIKILT heeft daarom een automatic linerexchange-systeem (ALEX), dat automatisch de glazen buisjes vervangt. Van der Lee: “We moeten na elke twintig monsters de liner vervangen, anders gaat dat ten koste van de scheiding.”
Data
Detectie met TOF-MS is volgens Gerssen de beste methode voor screening. “Het voordeel van TOF-MS is dat er vooraf niet duidelijk gespecificeerd hoeft te zijn naar welke componenten je kijkt zoals bij triple quad-MS. Als er een vraag komt over een component waar nog niet naar is gezocht, kan ik in mijn data kijken of deze stof aanwezig is. Triple quad-MS is over het algemeen gevoeliger, het nadeel is dat je meer doelgericht moet zoeken door vooraf moedermoleculen te selecteren.”
Nadelen zijn er ook. Door de brede analysetechniek geeft opslag van data op de
computer problemen, vertelt Gerssen. “De TOF genereert bergen data, en dan krijg je problemen met datamanagement. Waar sla je alles op en hoe houd je het overzichtelijk?” Van der Lee bevestigt dit probleem. “Elke 2D GC-run van een plantaardig monster duurt ongeveer een uur. We maken het maximaal aantal scans dat het apparaat aankan, dat zijn er tweehonderd per seconde. Van elke run krijgen we vierhonderd megabyte aan data.”
“De dataverwerking is de meest tijdrovende stap. In normale omstandigheden verwerken we gewoonlijk tien tot veertig monsters per week”, vertelt Van der Lee. “Het voordeel van onze apparatuur is wel dat we nu alle informatie uit één machine krijgen, waar we vroeger vijf verschillende apparaten nodig hadden voor dezelfde data.”
Ondanks alle hectiek blijft een goede verwerking van de data het belangrijkste. Traag: “Sommige analyses duren gewoon lang, dat kan niet anders. En als we iets rapporteren aan het ministerie, moeten we er 100 procent zeker van zijn dat de identiteit van de stoffen goed is vastgesteld en dat de kwantitatieve analyses in orde zijn. Je moet een bepaalde sfeer creëren waarin je de druk kan opvoeren, zonder dat je je mensen over de kling jaagt. Kwaliteit van de data staat altijd voorop, daar mag geen twijfel over zijn.”
