Hoe vind je in de zoektocht naar kanker een minuscuul percentage misvormde basen terug in een klein DNA-monster? De oplossing is overstappen op een minuscule kolom, meent de Antwerpse chromatografie-expert Eddy Esmans.
Zoeken naar de naald in de hooiberg, zo karakteriseert Eddy Esmans zijn stiel. En was het maar een hooiberg, want dan hoefde de Antwerpse hoogleraar zich minder te bekommeren over de grootte van zijn monsters. Wat hij in werkelijkheid zoekt is één gemodificeerde base op 100 miljoen, in de hoeveelheid DNA die valt te isoleren uit een rattenbeentje. “En da’s niet veel! Je kunt misschien 100 µg isoleren uit het weefsel, en daar moet je het mee doen.”
Vandaar dat hij ook de chromatografie heeft geminiaturiseerd. “Klassieke LC-kolommen met een diameter van 4,6 mm zijn niet gevoelig genoeg. Die hebben we uit het lab verbannen. We gebruiken alleen nog capillaire kolommen van 300 µm inwendig, of nanokolommen van 75 µm. En die koppelen we met nano-elektrospray-tandem-massaspectrometrie, een zeer gevoelige analytische techniek die zowel kwalitatieve als kwantitatieve informatie kan leveren.”
CARCINOGENEN
Esmans’ onderzoek spitst zich toe op DNA-adducten. “Die structuren ontstaan doordat bepaalde chemicaliën reageren en met DNA. Dat kan op twee manieren: óf de stof reageert als zodanig óf hij ondergaat eerst een metabole verandering, meestal in de lever. Een bekend voorbeeld is benzo(a)pyreen, een polyaromaat dat onder andere voorkomt in sigarettenrook en dieseluitstoot. Die stof is carcinogeen omdat het metaboliet, benzo(a)pyreen-7,8-diol-9,10-epoxide, met DNA reageert.” Hij legt uit dat de reacties meestal plaatshebben op de purine- of pyrimidinebase. “De adducten verstoren de waterstofbruggen tussen de double strands, of maken dat basen niet meer correct worden herkend. Er zijn ook stoffen die reageren op de fosfaatbackbone van het DNA, met het risico dat die breekt.”
Esmans voorspelt dat het binnen de industrie ooit routine zal worden om te controleren of een product geen kankerverwekkende adducten vormt. De nanokolommen die je voor zulke analyses nodig hebt, zijn nu al gewoon te koop. “De laatste jaren hebben de grote firma’s op deze miniaturiseringstendens ingespeeld. Eén van de eerste was LC Packings, later ook Waters en nog een paar.” De winst zit in de absolute gevoeligheid, benadrukt hij. “In vergelijking met een 4,6 mm kolom win ik een factor 3.700 aan gevoeligheid. Dat wil zeggen dat ik dezelfde concentratiegevoeligheid houd, maar bij een 3.700 maal kleiner monstervolume .Dit heeft zijn nut in combinatie met elektrospray ionisatie die zich gedraagt als een concentratiegevoelige detector.”
Daarmee is nog niet het probleem van de naald en de hooiberg opgelost. Als je een tot 2’-deoxynucleosiden enzymatisch gehydrolyseerd DNA-monster in een keer inspuit in de HPLC dan krijg je vier grote pieken van de ongewijzigde 2’-deoxynucleosiden. Maar waar zitten dan je adducten? “De oplossing heet column-switching. Voor de analytische LC-kolom zetten we een klein LC-kolommetje, van 5 mm lang, met een diameter van 500 of 300 µm. We vullen het zelf met een apolaire reversed phase-pakking. We injecteren een grote hoeveelheid, 20 µl of meer, van ons monster en we spoelen de kolom zo dat alle intacte 2’-deoxynucleosiden oplossen in de spoelvloeistof en alleen de gemodificeerde nucleosiden worden tegengehouden. Daarna veranderen we het solventsysteem dusdanig dat de adducten worden losgeweekt en naar de analytische kolom spoelen.”
DOWNSCALEN
Een rondgang door het lab aan de Groenenborgerlaan leert dat je er met alleen de kolommen nog lang niet bent. “Je moet alles downscalen. In plaats van een injectievolume van enkele microliters op een 4,6 mm kolom injecteer je theoretisch op een nanokolom slechts drie nanoliter. Dit is praktisch niet haalbaar, vandaar dat ook hier column-switching een oplossing biedt. En in plaats van een milliliter per minuut als debiet zit je bij een nanokolom op 200 nanoliter.”
Er zijn dus ook speciale pompen aangeschaft die op een accurate manier nanoliterdebieten kunnen leveren: de ‘klassieke’ LC-pompen zijn immers niet zo ver terug te regelen. Ook de elektrospray-interface van het MS-apparaat moet worden aangepast aan het lage nanoliterdebiet. “We gebruiken naast een nano-elektrosprayprobe ook zogenoemde PicoFrit-kolommen met een taps toelopend uiteinde, die je direct in de elektrospraybron kunt steken.”
Ondanks alle verfijningen is het allemaal prima herkenbaar voor iemand die een aantal jaren uit het vak is geweest. “Scheidingstechnologie wordt weer de kern van het onderzoeksdomein”, bevestigt postdoc ErwinWitters die zich in Esmans’ lab bezighoudt met proteomics. “De afgelopen vijftien tot twintig jaar is vooral onderzoek gedaan naar DNA. Nu zie je dat er wordt teruggegrepen op scheidingstechnologie om te ontdekken wat kleine moleculen en eiwitten doen.”
BORSTTUMOREN
Met de geminiaturiseerde apparatuur is in Antwerpen onder meer een borsttumoranalyse op punt gesteld. Statistische studies in de VS suggereren een verband tussen hormoonpreparaten voor vrouwen in de overgang, zoals premarine, en de kans op borstkanker. “Wat wij wilden zien, is of we met geminiaturiseerde elektrospray LC/MS adducten konden terugvinden in het DNA”, vertelt Esmans. “Het antwoord is ja. Van een achttal patiënten hebben we weefsel gekregen en we hebben daarin adducten kunnen vinden van equilenine, het hoofdbestanddeel van premarine.”
Een tweede toepassing betreft melphalan. “Dat is een bidentaat, met een structuur die lijkt op die van mosterdgas. Het wordt gebruikt voor de bestrijding van eierstokkanker. De werking wordt eraan toegeschreven dat de twee armen van het molecule een brug leggen tussen de strengen van het DNA zodat de deling niet meer doorgaat.”
Nadeel van melphalan is dat sommige patiënten na een jaar of negen secundaire (uitgezaaide) leukemie blijken te ontwikkelen. “Dus zijn we gaan zoeken naar adducten. We hebben ratten geïnjecteerd met melphalan en vervolgens weefsel geïsoleerd uit lever, nier en beenmerg. Het is gelukt om een standaard te identificeren, zodat we nu ook kwantitatieve proeven kunnen doen. Het grootste aantal adducten zit inderdaad in het beenmerg, en dat is een mogelijke verklaring voor die leukemie.”
Esmans voegt er aan toe dat hij van de vermeende crosslinking tussen de strengen tot nu toe geen enkel spoor heeft aangetroffen: “Dat heeft nogal geleid tot debat tussen mij en andere kankeronderzoekers.”
UNIEKE NICHE
Dertig jaar geleden kwam Esmans terecht in de chromatografie en organische massaspectrometrie toen zijn toenmalige patroon, prof. Alderweireldt, een van de eerste LC/MS-toestellen in België aanschafte. Nu, een paar maanden van zijn emeritaat verwijderd, staat het specialisme in Antwerpen stevig op de kaart.
“We proberen op allerlei gebieden een unieke niche te voeren, waar anderen geen zin, expertise of tijd voor hebben”, verklaart Esmans zijn strategie van zijn lab, dat minder personeel en apparatuur heeft dan bij grote bedrijven vaak het geval is. “Dan hoop je dat ze hierheen komen.”
