Vrijwel ieder laboratorium beschikt over GC’s en HPLC’s met een MS-detector. Zijn standaarden dan nog wel nodig ?
Standaarden zijn voor chromatografie even noodzakelijk als een scheidingskolom, een mobiele fase, een injector en een detector. Merkwaardig genoeg maken velen hun standaarden zelf, terwijl de overige attributen kant en klaar betrokken worden van een leverancier. Ook merkwaardig is dat alle GC- en HPLC-producenten ISO 9000 gecertificeerd zijn. Waarom eigenlijk, want als uw toestel onverhoopt temperatuur en druk foutief weergeeft, kunt u nog steeds uitstekende analyses doen. Het minste foutje echter in de standaard geeft onbetrouwbare antwoorden.
Een ISO 9000 certificaat geeft geen garantie voor de kwaliteit van het product, maar het geeft wel meer zekerheid dat, bij aanschaf van meer toestellen, ze allemaal even goed zijn of allemaal dezelfde afwijking hebben.
Voor gebruikers die hoge eisen stellen is dit een onbevredigende situatie. Voor hen is ISO 17025 ontworpen. Daarin staat aan welke criteria het toestel voldoet en op welke wijze dat getest is. Dit is nog steeds geen garantie dat het toestel het juiste antwoord geeft voor de door hen gewenste analyse.
Een bekend voorbeeld is de totaal-organo-halogeen analysator (EOX, TOX). Een in de handel zijnd gecertificeerde apparaat geeft bij deze stoffen soms slechts 30 procent van de juiste waarde. Een ander bekend toestel, dat niet gecertificeerd is, geeft ook voor hoogmoleculaire verbindingen betrouwbare waarden. Een certificaat op zich zegt dus niets, u moet goed lezen hoe het toestel getest is. Bij het gecertificeerde EOX-toestel wordt opgegeven dat het getest is met aldrin (volgens NEN 5735). Dit stofje geeft zelfs bij het slechtste toestel 100 procent opbrengst. Aldrin is derhalve geen maatstaf voor de kwaliteit van een EOX-toestel. Het is daarom een slechte keuze geweest van de betreffende NEN-commissie. De bouwwereld daarentegen die graag analyserapporten ziet met lage EOX-waarden, is blij met deze keuze.
Een ander voorbeeld is de testprocedure welke heeft geleid tot het heparine schandaal. Dit wereldwijd verspreid middel tegen bloedstolling is door de Chinese fabrikant illegaal bijgemengd met Chondroïtinesulfaat (OSCS), een veel goedkoper bloedstollingsmiddel, maar met ernstige bijverschijnselen. Dit werd vanwege de voorgeschreven testprocedure niet herkend. Een bonafide fabrikant is daarom belangrijker dan het te hoog inschatten van de waarde van een certificaat, waarin staat dat die standaard bij de productie de voorgeschreven eindcontrole goed heeft doorstaan. Twijfel aan de betrouwbaarheid van een gecertificeerd product blijft een schone zaak.
Het zal daarom duidelijk zijn dat ISO 9000 voor de fabricage van CRMs (certified reference materials) (standaarden en testmengsels) niet toereikend is. Daarin voorziet de reeds genoemde ISO 17025.
Deze norm eist een controle op de bereiding door middel van een vergelijkende chromatografische analyse. Maar dat niet alleen. Het bijbehorende certificaat geeft ook informatie omtrent de houdbaarheid van de standaard. Dit komt de betrouwbaarheid van het product ten goede. Men moet namelijk uitsluiten dat gedurende opslag reactie plaatsvindt met opgeloste lucht of dat de componenten onderling reageren.
Natuurlijk zal een vakkundige bereider van een mix-standaard geen componenten samenvoegen waarvan men op theoretische gronden een onderlinge reactie kan verwachten. Men dient desondanks het verloop van de samenstelling in de praktijk te controleren. In ISO Guide 35 staat beschreven hoe de CRM producer via een initial en on-going stability studies komt tot een expiration date van de standaard.
Als op een ISO 17025 certificaat een houdbaarheid van 3 jaar is vermeld, dan weet men dat een gelijke standaard al minstens drie jaar eerder is gemaakt en dat daarvan recent een chromatografische analyse is gedaan, waaruit blijkt dat de samenstelling niet veranderd is.
Als leverancier van CRMs werden we (auteur was 28 jaar directeur van Bester BV, groothandel in o.a. standaarden) regelmatig benaderd door klanten die een niet eerder geproduceerde custom made standaard hadden besteld en ontvangen. Sommige klanten lieten ons weten, dat de RvA (Dutch Accreditation Council) inspecteur daarmee geen genoegen neemt; hij eist dat op het certificaat een exacte houdbaarheidsdatum vermeld staat. Deze inspecteur begrijpt er dus niets van.
Hij behoort te weten dat een volgens ISO Guide 34 voor het eerst geproduceerde standaard als zodanig dient te worden gecertificeerd.
Gezien de machtspositie van het RvA weet de klant geen raad hoe aan hun eisen te voldoen. Het zou de RvA sieren als zij betere instructies geeft aan hun medewerkers.
Uit het volgende blijkt hoe verleidelijk het is verkeerd om te gaan met RCMs.
Stel: een 9000-2005 gecertificeerd lab koopt een gecertificeerde standaard met 15 componenten. Omdat deze seriematig wordt vervaardigd is dit voor de klant veel goedkoper en betrouwbaarder dan dat hij die zelf gaat maken. Maar voor zijn doeleinden had hij eigenlijk een standaard met 16 componenten nodig, welke helaas niet commercieel verkrijgbaar is. Hij mengt dan de zestiende component bij en verdeelt de standaard in veel ampullen, zodat hij een jaar vooruit kan. Hij beschrijft dit netjes in zijn managementsrapport en laat dat zien bij het eerstvolgend audit.
Wat gebeurt er: de inspecteur keurt de standaard af omdat de houdbaarheid van de gekochte CRM al gedeeltelijk verstreken is. Klant klaagt bij de leverancier en wil een standaard met langere houdbaarheid. Ook hier weer hebben we te maken met een verkeerde omgaan met gecertificeerde standaarden. De inspecteur had moeten zeggen, de gecertificeerde houdbaarheid geldt tot de datum dat men de ampul opent.
Gaat men daarna de samenstelling wijzigen, dan zal men een deel moeten achterhouden om later na te gaan of de standaard gedurende de gebruikstijd nog de verwachte samenstelling heeft. Men weet dus pas na een jaar dat de houdbaarheid van deze standaard een jaar is. Het is onzin om al bij de productie vast te stellen wat de gegarandeerde houdbaarheid is.
In ISO Guide 34 staat uitgebreid vermeld aan welke strenge eisen een reference material producer dient te voldoen. Hij zal een deel van de productie bewaren en na verloop van tijd de kwaliteit controleren. Zodoende krijgt hij een inzicht in de houdbaarheid. Ook kan hij daardoor eerder verstrekte standaarden hercertificeren en dit certificaat voorzien van een langere houdbaarheid dan vermeld staat op het oorspronkelijk certificaat. De bedenkingen hier tegen van sommige RvA inspecteurs zijn dus onterecht.
Als men dit alles overweegt moet men tot de conclusie komen dat het veel veiliger is de standaarden te betrekken van hierin gespecialiseerde leveranciers dan deze zelf te fabriceren. De specialist verkoopt de CRMs in grote aantallen en vaak wereldwijd. Mocht er een fout in het product zijn vanwege de toelevering of wat voor reden ook, dan is de kans dat ergens op aarde iemand de fout ontdekt, erg groot. ISO Guide 33 vermeldt zelfs, dat als een CRM erg duur is, dat als men goed kan rekenen, de CRM altijd nog goedkoper is dan zelf een standaard samenstellen.
Het hanteren en interpreteren van een certificaat is geen eenvoudige zaak. De laboratoriumscholen dienen hier voldoende aandacht aan te schenken.
Het RvA zal er goed aan doen meer te investeren in de opleiding van hun vakdeskundigen en van tijd tot tijd nascholing te verrichten. Alleen op die wijze kunnen de RvA-medewerkers één lijn trekken in hun optreden naar buiten.
